Dioplus 5/80mg (28 Tablet / Pack) is a fixed-dose combination oral antihypertensive medication designed for the comprehensive management of essential hypertension. It works via a synergistic dual-action pathway: amlodipine blocks transmembrane calcium influx into vascular smooth muscle to cause peripheral vasodilation, while valsartan selectively blocks angiotensin II type 1 receptors to inhibit systemic vasoconstriction and aldosterone secretion. This combined mechanism significantly lowers peripheral vascular resistance, decreases cardiac afterload, and controls elevated blood pressure.

Purpose

To treat essential hypertension in adult patients whose blood pressure is not adequately controlled on monotherapy regimens.

Warnings

Discontinue the medication immediately upon confirmation of pregnancy due to the risk of fetal toxicity and severe injury caused by ARBs. Can cause severe symptomatic hypotension or dizziness, particularly in volume-depleted, salt-restricted, or elderly individuals. Use with caution in patients with severe renal artery stenosis, congestive heart failure, or severe hepatic functional impairment. Monitor serum creatinine and potassium levels periodically, and avoid concurrent use of potassium supplements without medical advice.

Side Effects

Peripheral edema or localized fluid swelling around the ankles and lower feet due to amlodipine-induced vasodilation. Dizziness, lightheadedness, headache, or orthostatic hypotension upon changing postures from sitting to standing. Gastrointestinal symptoms such as mild nausea, dyspepsia, transient diarrhea, or temporary abdominal discomfort. Central nervous system reactions including somnolence, flushing, palpitations, or a feeling of general physical weakness.
