Naproxen is a non-selective non-steroidal anti-inflammatory drug (NSAID) with pronounced analgesic, antipyretic, and anti-inflammatory properties. It works via non-selective competitive inhibition of the cyclooxygenase enzymes 1 and 2 (COX-1 and COX-2). This inhibition blocks the bioconversion of arachidonic acid into pro-inflammatory prostaglandins. By reducing prostaglandin concentrations at peripheral tissue sites, it effectively diminishes the sensitivity of nociceptors to inflammatory mediators, limits localized tissue edema, and downregulates the hypothalamic thermoregulatory center.

Purpose

It is indicated for the management of rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, acute gouty arthritis, tendonitis, bursitis, and the management of primary dysmenorrhea and mild to moderate acute pain.

Warnings

**Gastrointestinal Risk:** Can cause severe gastrointestinal mucosal injury, ulceration, and occult gastrointestinal bleeding; use with extreme caution in patients with a history of peptic ulcer disease and consider concurrent cytoprotective therapy (PPIs). **Cardiovascular Warning:** May increase the risk of serious cardiovascular thrombotic events, myocardial infarction, and stroke; contraindicated for perioperative pain management in the setting of coronary artery bypass graft (CABG) surgery. **Renal Impairment:** Avoid use in patients with advanced or worsening renal disease due to the risk of acute kidney injury from reduced renal blood flow.

**Gastrointestinal:** Dyspepsia, epigastric pain, nausea, abdominal discomfort, heartburn, constipation, and occult bleeding. **Central Nervous System:** Headache, dizziness, somnolence, vertigo, and tinnitus. **Dermatological & Systemic:** Fluid retention, peripheral edema, pruritus, skin rashes, and ecchymoses.
